To a degree yes,beacuse your redrilling into the core.

Usually one plug and re-drill isn't that bad due to how minimal the amount of core removed is. If done correctly the reaction will be different but in a good way if being done to change lay out. If just moving a thumb everything is pretty minimal.

Once It would only affect it to minimal degree. But The more you do it, the more you could change it.
